The method of preparation is about the subtlety. Today there is a buffer that combines Tris and acetic acid, and its pH range is the key to making this substance.

To make this buffer, the ratio of Tris to acetic acid must be carefully reviewed. If you want to get a buffer in a specific pH range, you should calculate the amount of both according to chemical reasons. If you want to make the pH more acidic, you should increase the amount of acetic acid slightly; if you want to be close to alkaline, you should add Tris.

When preparing, first take an appropriate amount of Tris and put it in a clean container, dissolve it in water, and stir slowly to make it completely soluble. Then according to the calculated amount, slowly add acetic acid, and continue to stir between them to ensure that the two are evenly miscible. During the process, a precision pH meter must be used to monitor the pH value of the solution in real time, and the amount of acetic acid or Tris should be fine-tuned according to the gap between it and the target pH range.

When the pH value is just within the preset range, this buffer will be prepared. After it is made, it should not be slack, and it should be properly stored. It should be placed in a cool, dry place, and tightly sealed to prevent it from mixing with impurities in the air, causing pH range variation and losing its original effect.
