Then, the spotting work also needs to be accurate. Dip the sample in a micro-applicator and carefully point on the starting line of the film. The amount of spotting is related to the clarity of the strip and the effectiveness of separation. If the amount is too large, the strips are prone to tailing and overlapping; if the amount is too small, the signal is weak and difficult to interpret accurately.
Then, place the spotted film in the electrophoresis tank to ensure that the film is properly connected to the electrode to ensure smooth current conduction. According to the characteristics of the sample and the purpose of analysis, set the appropriate voltage and current to be long in time. Generally speaking, the voltage is maintained within a certain range, which not only ensures the effective migration of the sample, but also avoids the risk of sample denaturation due to overheating.
After electrophoresis, the film needs to be dyed to reveal the sample bands. Common stains vary depending on the type of sample, and amino black is often selected for proteins. After dyeing, the excess dye solution is rinsed and removed, and the strips are clearly presented. According to the position, width and color depth of the strips, the sample composition, purity and other information can be analyzed.
