Preparation of Ammonium Acetate Buffer (pH 4.5)
To prepare ammonium acetate buffer, so that its pH value reaches 4.5. An appropriate amount of ammonium acetate and acetic acid should be prepared.
First take a certain amount of ammonium acetate, dissolve it in an appropriate amount of pure water, stir until it is completely dissolved, and obtain an ammonium acetate solution.
Then slowly add acetic acid to the above solution, and measure its pH value with precise pH. Be careful during the dripping process and pay close attention to the change of the pH meter.
When the pH value is gradually approaching 4.5 and approaching the target value, it is even more necessary to add acetic acid dropwise, and strive to adjust it accurately. When the pH value is just 4.5, stop dripping.
In this way, the ammonium acetate buffer (pH 4.5) is prepared. During the preparation process, the utensils used should be clean and the operation should be standardized to ensure the quality and accuracy of the buffer.
