Preparation of 100 mmol ammonium acetate buffer
To prepare 100 mM ammonium acetate buffer, the following method is required.
Prepare an appropriate amount of ammonium acetate solid and ultrapure water first. Accurately weigh a certain amount of ammonium acetate. Calculate according to the formula. If V liters of 100 mM ammonium acetate buffer are prepared, and the molar mass of ammonium acetate is M (g/mol), the required ammonium acetate mass m = 0.1 mol/L × V L × M g/mol. After weighing, place it in a clean beaker.
Slowly add an appropriate amount of ultrapure water to the beaker, and stir at a constant speed with a glass rod to promote its dissolution. After the ammonium acetate is completely dissolved, transfer the solution to a suitable volume flask. Rinse the beaker and glass rod several times with a small amount of ultrapure water, and the rinsing liquid is also moved into the volumetric flask to ensure that there is no loss of solute.
After that, use ultrapure water bandwidth evaluation to the scale line of the volumetric flask, cover the cork, and shake it upside down to make the solution uniform.
If you need to adjust the pH of the buffer, you can add it dropwise with dilute acetic acid or dilute ammonia water according to actual needs, and accurately monitor it with a pH meter until the desired pH is reached. In this way, 100 mM ammonium acetate buffer is prepared.
